Is there a way to increase the line spacing when the font for buttons and labels is increased? On my 4K monitor these fonts are very small. I can increase to 13pt (which is still tiny), but the line spacing stays the same, so any further increase and the fonts are obscured.

Not possible within XY. You should use the global text scaling provided by Windows display settings instead.
