Enabled - on the icon only,and single click to open an item doesn't work sometimes,I have to click 2 or 3 times then only it opens.

There could be a conflict with MDBU. Try to turn off this: Configuration | Preview | Mouse Down Blow Up | Mouse Down on Thumbnails and Icons | On left mouse down

that has solved it,but how to use Mdbu with left mouse click and at the same time single click to open an item.

somehow make both work admin,is it not possible

I have those same settings and single clicking an icon to open a file and MDBU both work fine. What is the setting of Configuration > Mouse Down Blow up > Allow dragging by thumbnail. If that's selected, try turning unselecting it. In case you didn't know, that setting also slows down MDBU cause XY has to take a short amount of time to figure out what you're going to do, MDBU or drag.
