I feel you… It only occurred to me a few years ago (never since), but it was definitely frustrating!
I don’t think what you ask for is implemented as is by default (wouldn’t be so wise for various reasons).
But here’s what you could do instead:
☛ General suggestion: Assign a shortcut to "Save Settings"
Shift+F9 > Jump… > type "save" > OK >
Assign something logical for you, e.g. "Ctrl+S"
Not exactly what you were after, but probably enough—and a sensible action anyway IMHO!
☛ Specific suggestion: Assign shortcuts to "Label # and Save Tags (& Labels) Database"
Menu "User" | "Manage Commands…" > Choose "Run Script" in left pane >
"New…" (add an entry for each of your labels)
e.g. for the first label:
Caption: Apply Label 1
Script: tag 1; #195;
Assign Keyboard Shortcut…: e.g. Numpad 1
FYI, I personally assigned
Numpad 1-5 to labels 1-5
Numpad 0 to "Remove Label" (tag 0;), and
Numpad . to "Apply Last Label" (#879;)
(and also Num6 to "Add Comment…" and Num7 to "Add Tags By List…", etc.)
(Matching #numbers are in "Help" | "List All Commands…")
I can tell you it’s a breeze to label items this fast (if you have a pad, ofc)!
And BTW, I only save manually with Ctrl+S after extensive labeling sessions (don’t need to force save after each individual label).

Hm - maybe it's not exact about "settings"?
I think it's more like an attribute of a file - that should be saved when changed. Independent from XY-settings, being something like XY-internal.

I’d overlooked this info—drawing from own experience, where occasionally I’d rather save everything on demand (including e.g. User Scripts, etc.), and didn’t have to change this workflow since labels were introduced.

I just wondering why not a immediately effect, like changing the filename.
OSX Finder's label is immediately saved.
now I assign "Ctrl + S" to Save Tags (labels, tags and comments), but I really hope that this process can be automated.

- Saving a potentially huge file on every single tag changing mouse click or other action is simply too much.
- It's also a good safety belt to have a good version on file in case of a fatal slip of the hand.
- The Autosave settings after this many minutes setting does a good job enough here.
